molecularNote  An insertion vector composed of a floxed lacZ and polyadenylation signal followed by a GFP tagged human histone H2B and, finally, an FRT flanked neomycin selection cassette was inserted 6 bp upstream of the ATG start site in exon 1. Expression of flp recombinase in properly targeted cells removed the selection cassette. Ecre excision then removed the flozed lacZ permitting expression of the tagged histone.
