molecularNote | This transgene contains the human calcium/calmodulin-dependent protein kinase II alpha (CAMK2A) promoter driving expression of a cDNA encoding human Kv channel interacting protein 3, calsenilin (KCNIP3; also called downstream regulatory element antagonist modulator (DREAM)) with two amino acid substitutions each at EF hands 2, 3 and 4 (EFmDREAM). The cDNA is followed by an internal ribosomal entry site (IRES) and a lacZ reporter gene. The amino acid substitutions render the mutant protein insensitive to Ca2+; it thus can block the Ca2+-dependent derepression function of endogenously expressed KCNIP3 and other Kv channel-interacting proteins. Line 26 is reported to exhibit "higher and clearer expression" in the telencephalon than line 1 or line 33. Distribution in line 26 is reported to be restricted to telencephalic neurons with trigeminal neurons devoid of noticeable expression. |
