molecularNote | The cDNA sequence encoding mouse C1q and tumor necrosis factor related protein 3 (C1QTNF3; also called CTPR3) with a carboxy-terminal FLAG epitope (DYKDDDDK) tag was cloned into the pCAGGS vector downstream of the ubiquitously expressing "CAG promoter" - comprising the cytomegalovirus immediate early (CMV-EI) enhancer followed by the promoter, first exon and first intron of the chicken beta-actin gene with the 3' splice junction sequence replaced by that of the rabbit hemoglobin beta gene - and followed by the rabbit beta-hemoglobin polyadenylation signal. At the mRNA level, expression is observed in all tissues of transgenic mice analyzed by RT-PCR. The FLAG-tagged protein is detected by immunoblot analysis in heart, kidney, liver, and muscle, but not in adipose tissue or brain. Mice of the single maintained transgenic line exhibit greater than fivefold the wild-type level of circulating C1QTNF3 protein. |
