| molecularNote | The 12-kb transgene, derived from the pPrHG "half genomic" vector, comprises the promoter and first exon of the mouse prion protein gene (Prnp); mouse exon 2 joined to exon 3, containing the entire prion protein open reading frame, from kudu (an antelope of the genus Tragelaphus); and the 3'-untranslated region (UTR) of the mouse gene. The encoded protein contains six copies of the amino-terminal octapeptide repeat sequence (6OR), rather than the usual five copies. Mice bearing this transgene express kudu 6OR prion protein from the mouse Prnp promoter in brain and other tissues. |
