| molecularNote | A sequence was engineered comprising the tdTomato red fluorescent protein open reading frame followed by the SV40 polyadenylation sequence and an FRT site-flanked neomycin resistance cassette. This was inserted into the BAC RP23-407K8 immediately downstream of the translation-initiating ATG in exon 1 of the mouse glutamic acid decarboxylase 2 gene (Gad2; also called Gad65); the BAC contains all 16 exons, 72 kb of 5'- and 64 kb of 3'-flanking DNA. The neo cassette was removed by flp-mediated recombination. Of 11 founder mouse lines that had integrated the transgene and that transmitted it through the germline, only one exhibited the expected expression pattern upon fluorescence imaging of brain sections. |
