molecularNote | In male mice with this transgene, expression of cDNA encoding the mouse kidney androgen regulated protein (KAP) is driven by the mouse Kap promoter and the distal regulatory sequences of the human angiotensinogen gene (AGT), which together confer androgen responsiveness. The Kap cDNA was cloned downstream of the Kap promoter (nucleotides -1542 through -1 relative to the transcription initiation site) into the NotI site of the pKAP2 vector. This is followed by the last 42 bp of human AGT exon 2 through exon 5, which lost their coding potential owing to the lack of the first 817 bp of exon 2 that included the ATG start site. RT-PCR and western blot analysis of samples from various tissues detects no expression in tissues other than kidney and demonstrates approximately twice the level of both mRNA and protein in kidneys of 6-8 month old male transgenic vs. control mice. Immunohistochemical analysis using an anti-KAP monoclonal antibody showed the protein to be restricted to the proximal tubule cells of the kidney. However, at higher magnification "diffuse, intense staining" was apparent throughout transgenic male mice; control males exhibited less intense general staining. |
