| molecularNote | A cassette bearing a tdTomato/T2A/Cre-ERT2/bGH polyA was inserted in frame with the mouse Mex3a start codon in exon 1. After homologous recomibination in ES cells, positive clones were expanded and then transfected with an FlpO-bearing plasmid to remove the neomycin cassette. Positive clones were selected and blastocyst injection was done to generate chimeras. The chimeras with successful germline transmission were bred with C57BL6 mice and the offspring was used to generate the final mouse colonies. |
