molecularNote | The ~180.5 kbp C57BL/6J mouse bacterial artificial chromosome (BAC) RP23-247J9 was obtained; containing the entire surfactant associated protein C locus (Sftpc) as well as ~70 kbp of centromeric flanking sequences (including the complete loci Phyhip and Bmp1) and ~108 kbp of telomeric flanking sequences (including the complete loci Lgi3, Reep4, Hr, Nudt18, Fam160b2 and Nudt18, as well as a 3' portion of Dmtn). Using homologous recombination/BAC recombineering, a 2230 bp GCaMP8-SV40pA construct was inserted into the ATG start site of the BAC Sftpc gene. No other loci on the BAC were altered. The 11.6 kbp BAC vector (pBACe3.6) contained a chloramphenicol selection cassette. The genetically encoded calcium indicator GCaMP8 is a sensitive GCaMP variant with high dynamic range and fast response kinetics. Founder line B23-5 was identified with high sensor expression in pulmonary type II alveolar epithelial cells. |
