molecularNote | A ~5.7 kb genomic fragment, containing the Myh6 (myosin, heavy polypeptide 6, cardiac muscle, alpha) regulatory region, was used to drive the cardiac muscle expression of the bacterial chloramphenicol acetyl-transferase reporter gene. The genomic fragment contains sequence from a BamHI restriction site in the last intron of the neighboring Myh7 (myosin, heavy polypeptide 7, cardiac muscle, beta) gene to the MaeIII site at 1074 in Myh6 exon 3. It contains the following elements: ~100 bp of the last intron plus the entire last exon of Myh7, ~4 kb of intergenic sequence (containing CCAAT and TAATA boxes and other promoter sequences), and the 5' end of the Myh6 gene (comprising the first two (5' UTR) exons and introns, and most of the the non-coding part of exon 3). Following micro-injection of the transgene, two germ lines were established (7 and 27). Chloramphenicol acetylation activity was only detected in the heart and not in aorta, diaphragm, breast, biceps, adductor magnus, gastrocnemius, kidney, liver, brain, uterus, thigh, calf or spleen. There was very low expression in lung. |
