| molecularNote | A targeting vector was designed to replace the Stk33 locus between exon 6 and exon 8 with a cassette containing exon 7 of Stk33, a neomycin resistance gene flanked by FRT sites, and loxP sites surrounding exon 7 and the neomycin cassette. The mice that still contained the neomycin cassette were bred with CMV-Cre mice leading to germline deletion of Stk33 exon 7 together with the neomycin cassette creating the straight Stk33 knockout mouse. The deletion of exon 7 removes part of the Stk33 kinase domain and leads to a frameshift resulting in a premature stop codon at the second amino acid in exon 8. Immunohistochemistry and western blot confirmed absence of protein in testis, however shortened proten lucking 76 amino acids of the kinase domain including the active site coded by exon 7 and exon 8 were present in the lung and oviduct. |
