A vector containing an optimized Kozak translation initiation sequence, a beta-globin poly-adenylation signal (BpA), the mApple fluorescent reporter, and an FRT-flanked kanamycin/neomycin resistance cassette is inserted into the start codon of the Glp1r gene within the BAC clone. The FRT-flanked cassettes were removed by transient transfection of a FLP recombinase-expressing plasmid. |
