| molecularNote | This transgene construct contains the following elements: mouse myosin heavy chain 6 promoter sequence (also contains last exon of Myh7 and the first three, 5' UTR containing, exons of Myh6) the human myosin heavy chain 7B cDNA (excluding sequence for human-specific 42 aa extension, and with the sequence for the last 10 aa (RDALGPKHKE) replaced with a c-Myc tag (EQKLISEEDL) and human growth hormone gene poly(A) sequences. This allele drives the expression of the human gene in cardiomyocytes, where the Myh7b peptide is not normally expressed. Of the two founder mice, the mouse with the highest expression (~30% of total cardiac myosin) was chosen for further studies. |
