| molecularNote | A construct consisting of a bioluminescence synthetic Renilla luciferase (hrl) reporter gene, a reporter gene encoding the monomeric red fluorescence protein (mrfp1), and a mutant herpes simplex virus type 1 sr39 thymidine kinase [HSV1-truncated sr39tk (ttk) and a PET reporter gene ("3MR") was inserted in-frame with Cdkn2a exon 2 in the mouse BAC clone RP24-322D20. Expression of this construct results a fusion protein containing the first 62 amino acids of the encoded protein and the 3MR reporters, but no full-length WT protein. Insertion of the 3MR cDNA also resulted in a stop codon in exon 2 to prevent expression from the BAC gene. This transgene is designed to identify, isolate, and selectively kill senescent cells. |
