| molecularNote | A minigene containing floxed Vav1 exon 25-27 and FRT-flanked neomycin selection cassette were introduced in intron 24 and the downstream exons 25 and 26 were replaced with Vav1 exons 25-27 fused to the cDNA sequence corresponding to exons 25-28 of Myo1f. Exon 24 of Vav1 is spliced into the exon 25-27 minigene resulting in expression of a wild-type Vav1 mRNA. The neomycin cassette was removed via flp-mediated recombination. Cre recombinase expression is needed to remove the floxed exon 25-27 minigene to lead to exon 24 being spliced into the Vav1 exon 25-27Myo1f exons 25-28 minigene, which will result in the expression of the Vav1-Myo1f oncogenic fusion. |
