| molecularNote | An IRES-Tau (Mapt)-lacZ cassette was introduced immediately downstream of the exon 2 splice acceptor site and exons 2-4 and part of exon 5 were replaced by an FRT-flanked selection cassette consisting of a flp recombinase gene under the control of the testis specific promoter of the angiotensin converting enzyme gene and the neomycin resistance gene. Self-excision of the selection cassette was not seen so it was removed via flp-mediated recombination. A point mutation was identified in the lacZ gene such that beta-galactosidase expression is not observed. Western blot analysis and immunofluorescence in homozygotes confirmed absence of protein in neocortex and hippocampus, and in the retina, respectively. |
