molecularNote  The targeting vector was designed with (from 5' to 3') a ubiquitously expressed CAG promoter, a loxP-flanked stop cassette, a reverse tetracycline-regulated transactivator (rtTA3) sequence, P2A self-cleaving sequence that mediates ribosomal skipping, a mutant form of GFP, a T2A self-cleaving sequence, and a mutant form of Luciferase (luc). A frt-flanked blue fluorescence protein (eBFP2) sequence was inserted downstream. Also, a neomycin resistance (neo) cassette is present in the floxed region. This targeting vector was inserted into the Gt(ROSA)26Sor locus. Specifically, the targeting vector contains antigen-intact but non-functional forms of GFP (Delta T64) and luc (G315A). FLP recombinase removed eBFP2 cassette. Cre-recombinase removed the loxP-flanked stop cassette.
