| molecularNote | This allele was created using the FLEx switch system. A loxP site was inserted upstream of exon 10, a lox511 site, inverted exon 10 containing a valine to tyrosine substitution at amino acid 338 (V338Y), an FRT flanked neomycin selection cassette, loxP site in reverse orientation, and a lox511 site in reverse orientation were inserted downstream of exon 10. The neomycin cassette was removed via flp-mediated recombination. Cre-mediated recombination in the germline removed the wild-type exon 10 and flipped the duplicated inverted exon bearing the V338Y mutation resulting in expression of the mutant allele. Overall expression level of mutant mRNA in the brain, inner ear and retina of homozygous mutant mice is comparable to that in wild-type mice, indicating no deregulation of Mrps5 gene expression. |
