abstractText | We established a stable mouse myoblast cell line C2C12 in which human myotonin protein kinase (MtPK) cDNA is overexpressed. Total RNA was recovered from those and the control C2C12 cells and cDNA was synthesized by reverse transcription. Using differential display analysis, we cloned a novel gene that is enhanced under the transcriptional activation of MtPK. Northern analysis of mouse tissues revealed that the novel gene is ubiquitously expressed. On the other hand, a slight decrease in gene expression was observed in human DM samples in which MtPK expression was suppressed. The subcellular localization of the product is in the cytosol, but it degrades rapidly after transfection. When overexpressed in COS-cells, a granular fluorescene was observed in the cytosol and nucleolus. We named this gene product Dystrophia Myotonica- related protein (DMRP). |
