abstractText | Full text of MNL contribution: Identification and mapping of new loci using cloned cDNA. I have started to screen the mouse genome for mutations at the DNA level. A library of cDNA-containing plasmids has been made by Drs . K. Gross, F. Berger, G. Watson, and K. Paigen. They used a size fraction of mouse kidney mRNA, inserted the cDNA fragments into the Pst-1 site of pBR322 by C/G tailing and transfected into E. coli X 1776. Labelled recombinant plasmid DNA from several isolated clones has been hybridized to restricted electrophoresed DNA isolated from mouse livers. The sizes of restriction fragments which hybridize to labelled recombinant cDNA have been compared using different mouse strains . Several polymorphisms involving differences in fragment sizes have been detected in this way. Preliminary mapping using RI strains has revealed loci on chromosomes 4, 7 and 14, as shown. K); Locus: RP6; Chromosome: 7 At a discussion on nomenclature at the Biochemical Genetics Workshop held at the Jackson Laboratory in September 1980 the following guidelines were suggested. Loci should be named using letters derived from the institution at which the work took place. They should be numbered serially. Both letters should be capitalized t o indicate that the locus is at the level of the DNA, detected by recombinant DNA techniques. The locus name should be temporary, being replaced by a name indicating the function of the locus when known. Allele designations were left open. The meeting included Ms. Pricilla Lane, Ms. Muriel Davisson, Dr. Thomas Roderick and Dr. Margaret Green of the Jackson Laboratory, Dr. Muriel Nesbitt of the University of California , San Diego and Dr. Rosemary Elliott of Roswell Park Memorial Institute, Buffalo, N.Y.
