Povey, 1MRC Radiobiology Unit, Harwell, and 2MRC Human Biochemical Genetics Unit, University College London, London The gene for biliverdin reductase (BLVR; E.C. 1.3.1.24) has recently been assigned to the p14-->cen region of human chromosome 7 (Meera Khan et al., Biochem. Genet. 21:123, 1983; Parkar et al., Ann. Hum. Genet., in press, 1983). The genes for malate dehydrogenase NAD (mitochondrial), MDH2 and beta-glucuronidase, GUSB, have been assigned to the p22-->q22 and cen-->q22 regions respectively of human chromosome 7. In the mouse, the homologous genes, Mor-1 and Gus are located in the distal half (E2-->ter) of chromosome 5. Thus, there is a region of homology within human 7p22-->7q22 and a part of mouse chromosome 5. The finding of an electrophoretically detectable variant of biliverdin reductase in the mouse has enabled us to determine if the homologus gene, Blvr, is also on chromosome 5. It is not. No evidence of linkage was found between Blvr and three markers, sash, Wsh, angora, go, and buff, bf, on mouse chromosome 5 in backcross tests. Further linkage studies have shown that Blvr is linked to the coat color marker, non-agouti, a, on chromosome 2, for in 52 mice arising from the backcross Blvra a/Blvrb + female x Blvra a/Blvra a male 21 Blvra a/Blvra a, 24 Blvrb +/Blvra a, 3 Blvra +/Blvra a and 4 Blvrb a/Blvra a mice were found. The recombination percentage between Blvr and a is 13.5 +/- 4.7%. These data indicate that the region of homology within human chromosome 7 and mouse chromosome 5 does not include BLVR. It seems probable that the conserved segment lies within 7p14-->7q22, not 7p22-->7q22 and that MDH2 will be located in this region. Furthermore, it is expected that BLVR will be distal to MDH2.
