abstractText  Full text of MNL contribution: 7. MOUSE GENE MAPPING IN SOMATIC CELL HYBRIDS. We have employed Chinese hamster x mouse somatic cell hybrids segregating mouse chromosomes to map genes coding for enzymes which are not variant among inbred mouse strains. This work is part of a comparative gene mapping study designed to determine the conservation or disruption of linkage groups of homologous genes between man and mouse. Information from these studies will aid in our understanding of the evolution of the mammalian genome [Lalley et al., PNAS 75: 2382 (1978) and Lalley et al., Nature 274: 160 (1978)]. Using somatic cell hybrids it was shown that the genes coding for sorbitol dehydrogenase (SDH) and the tissue form of acid phosphatase (ACP-2) are located on mouse chromosome 2. In man these genes are located on chromosome 15 and 11, respectively, indicating that these linkage groups are not conserved. In collaboration with J.D. Minna and Uta Francke the gene coding for alpha-fucosidase (FUCA) was shown to be on mouse chromosome 4 further demonstrating the conservation of a large linkage group consisting of six genes located on mouse chromosome 4 and the short arm of human chromosome 1 [Lalley et a1., PNAS 75: 2382 (1978)]. In man genes coding for aminocylase (ACY) and Beta-galactosidase-1 (GLB1); phosphoglucomutase-2 (PGM2) (homologous to mouse PGM-1) and peptidase S (PEPS) (homologous to PEP-7); and malate dehydrogenase-2 (MDH2) (homologous to MOR-1), Beta-glucuronidase (GUS) and argininosuccinate lyase (ASL) are assigned to the short arm of human chromosome 3, 4, and 7, respectively. In the mouse Beta-galactosidase is located on chromosome 9 and Pgm-1, Pep-7, Mor-1 and Gus are on chromosome 5. In collaboration with S. Naylor and T.B. Shows, we demonstrated that Asl and Acy are located on mouse chromosome 5 and 9, respectively. Thus, the linkage of Asl - Gus - Mor-1, Acy - Bgl and Pgm-2 - Pep-7 are conserved between man and mouse suggesting chromosomal homologies between the proximal ha1f of mouse 5 and the short arm of human 4, the distal half of mouse 5 and the short arm of human 7, and a region of mouse 9 and the short arm of human 3. In collaboration with G. Koch and T.B. Shows, we have also demonstrated that the gene coding for acid lipase A (LIPA) is located on mouse chromosome 19. These data would indicate the linkage of the genes coding for LIPA and GOT-1 is also conserved in man and mouse. The data are summarized below. Enzyme: Acid lipase; Mouse Chromosome: 19; Proposed Symbol: Lipa; Conserved Linkage Group: Lipa - Got-1; Human Chromosome: 10. *Afuc has already been reserved as symbol for this locus (Ed.) The proposed nomenclature was chosen to conform with that used for the homologous genes which already have been mapped in man except for sorbitol dehydrogenase. The human gene symbol for this enzyme is SORD. The other differences in mouse and human gene nomenclature in the above table are: Pgm-2 = PGMl; Gdp-1 = GDH; Mor-1 = MDH2 and Bgl = GLBl.
