abstractText | Full text of MNL contribution: Electrophoretic variation for alcohol dehydrogenase ADH-1. An electrophoretic variant of liver and kidney alcohol dehydrogenase has been found in feral M.m. musculus trapped in Skive, Denmark, and supplied to us by Tonnes Nielsen of Aarhus University. Electrophoretic polymorphism for this enzyme was originally reported in feral mice trapped in Denmark by Selander, Hunt and Yang (Evolution 23, 379; 1969). Inbred strains are monomorphic for a cathodally migrating form (ADH-1B) in starch gels, pH 7.0. The variant, ADH-1A, is slower migrating, more negatively charged, than the inbred form. ADH-1AB heterozygotes have a three banded pattern consistent with a dimeric enzyme structure. Backcrosses of ADH-1AB with the inbred strain C3H/ST segregated 19 ADH-1AB: 20 ADH-1B which is consistent with a 1:1 ratio of a single gene. Similar ratios were observed in several crosses. We propose the name alcohol dehydrogenase-1 for this locus with the symbol Adh-1. The common inbred strains would have the allele Adh-lb and the new variant would be Adh-la. We have tested linkage of Adh-1 with loci shown in the following table: locus: Dip-1; chromosome: 1; % recombination: 55 locus: Id-1; chromosome: 1; % recombination: 60 locus: a; chromosome: 2; % recombination: 44 locus: Gpd-1; chromosome: 4; % recombination: 43 locus: Pgm-2; chromosome: 4; % recombination: 56 locus: Gus; chromosome: 5; % recombination: 52 locus: le; chromosome: 5; % recombination: 53 locus: Pgm-1; chromosome: 5; % recombination: 54 locus: Gpi-1; chromosome: 7; % recombination: 63 locus: Es-2; chromosome: 8; % recombination: 50 locus: Mod-1; chromosome: 9; % recombination: 44 locus: Mpi-1; chromosome: 9; % recombination: 46 locus: Es-10; chromosome: 14; % recombination: 54 locus: Pgk-2; chromosome: 17; % recombination: 44 No significant linkage relationships were observed. A homozygous Adh-1a stock is available to send to other laboratories. (Chapman, Blaird, Murawski, Quarantillo) |
