abstractText | Full text of Abstract: SOMATIC CELL GENETICS. CYTOLOGICAL LOCALIZATION OF THREE GENES ON CHROMOSOME 14 IN MUS MUSCULUS. L.C. Samuelson, and R.A. Farber. Univ. of Chicago, Chicago, Illinois. Using somatic cell genetic mapping techniques, we determined the regional map location of the mouse adenosine kinase gene (Adk) in relation to the genes for nucleoside phosphorylase (Np-1) and esterase-10 (Es-10). Cytological localization of these genes on mouse chromosome 14 was made possible by analysis of Chinese hamster-mouse hybrid cell lines which had been gamma-irradiated to induce mouse chromosomal rearrangements. Irradiated clones were screened for expression of mouse Adk, Np-1, and Es-10; segregant clones which expressed only one or two of these three markers were karyotyped. We observed several different rearrangements of mouse chromosome 14 in the segregant lines. Including deletions, translocations, and one inversion. The patterns of Adk, Np-1, and Es-10 expression in these lines were compared with the cytologically characterized rearrangements so that these genes could be mapped to specific Giemsa-band regions on chromosome 14. The order of these three genes was determined to be centromere-Adk-Np-1-Es-10. |
