The murine rd mutation is inherited as an autosomal recessive which results in the total degeneration of retinal photoreceptor cells by 4 weeks of life. While the biochemical basis for the rd phenotype has not been determined, it is known that abnormally high levels of cGMP occur in the rd retina a few days before the degeneration begins, and that this cGMP elevation is due to a deficiency in cGMP-phosphodiesterase (cGMP-PDE) activity. It is also known that the rd mutation maps to mouse chromosome 5. Therefore, we have determined the chromosomal location of genes for a number of visual proteins that could affect the activity of cGMP-PDE. Southern blot hybridization of DNAs from hamster x mouse somatic cell hybrids showed that the gene for the alpha subunit of transducin (Gnat-1) maps to chromosome 9, retinal S-antigen (Sag) to chromosome 1, interphotoreceptor retinoid binding protein (Rbp-3) to chromosome 14, the gamma subunit of retinal cGMP-PDE (Pdeg) to chromosome 11, and rhodopsin (Rho) to chromosome 6. Analysis of DNAs from an interspecific backcross has provided more specific map locations for several of these genes. Sequences homologous to cDNA for the Beta subunit of transducin were mapped to chromosome 4 (Gnb-1), chromosome 5 (Gnb-2), and chromosome 8 (Gnb-3) using cell hybrids. Analysis of the interspecific backcross showed that the only gene identified in this study on chromosome 5, Gnb-2 does not map near the rd mutation.
