abstractText  Full text of MNL contribution: New Mutations and Biochemical Variants. Work done in collaboration with Dr. Roderick, Dr. Taylor, Dr. Womack, and Ms. Lisa Hirshhorn (precollege student) of the Jackson Laboratory. Genetic variation in alkaline phosphatase isozymes has been detected in liver, kidney, and intestine, and the existence of four new loci is apparent on the basis of strain distribution. Their description follows. 1. A fast migrating isozyme requiring manganese is found in liver and kidney. Gene symbols proposed are Akp-la and Akp-lb for the fast and slow migrating forms. Eight of 30 recombinant inbred strains showed recombination with the Mls locus, placing Akp-1 on chromosome 1. Combined data from a two and a three point backcross revealed the following recombinants: Akp-1 and Lp 13/136; Akp-1 and Dip-1 32/105; Lp and Dip-1 20/105 which gives the following gene order and recombination frequencies: Akp-1 9.6 +/- 2.5%; Lp 19.0 +/- 3.8%; Dip-1 There were three double recombinants. This places Akp-1 more distal to the centromere than all other known loci on chromosome 1. A second isozyme requiring manganese was also present in liver but was not variant among the strains tested.
