INTRODUCTION Secretogranin 1 (chromogranin B) and carboxypeptidase H are secretory granule proteins involved in processing of peptide hormones in endocrine cells (e.g. insulin processing in pancreatic Beta cells). During our studies of the genetic predisposition for spontaneous, insulin-dependent diabetes in the nonobese diabetic (NOD) mouse strain, we tested for potential association of the structural genes Scg-1 and Cph-1 with the predisposition for diabetes in this strain. In the course of this study, we determined chromosomal localizations of both genes in the mouse. MATERIALS AND METHODS The probes used were of rat origin. The Scg-1 clone is a cDNA fragment encompassing 600 base pair (bp) of the 3' end of the transcript isolated from rat insulinoma (RIN) cDNA library. The Cph-1 clone (DG-1) is a 408 bp cDNA fragment isolated from a rat pancreas cDNA library. Both cDNAs are cloned in pBLUESCRIPT. Southern blot analyses were performed as described (1) and final washes after hybridization were at 60 degrees C in 0.l x SSC, 0.1% SDS. RESULTS Southern blot analysis of restriction fragment length variants (RFLVS) identified two Scg-1 alleles and three Cph-1 alleles (Table 1). Combined BC1 and AKXD RI analysis positioned Scg-1 within 8 cM from Il-1a on Chr 2 (Table 2A). Strain distribution patterns (SDP) of Cph-1 alleles in BXD, BXH, CXB, AXB, and BXA RI showed close concordance with Nat-1, Lpl, and Es-1 markers encompassing approximately 13 cM segment on Chr 8 [(2), Tab. 2B]. The best interpretation of these RI data is consistent with a position of Cph-1 between Nat-1 and Lpl. Preliminary results did not indicate a linkage of Scg-1 or Cph-1 with diabetes susceptibility (not shown). DISCUSSION Using rat cDNA clones for Scg-1 and Cph-1, we mapped the mouse homologues to Chr 2 and 8, respectively. Both probes identify RFLVs among inbred strains and the loci provide useful markers for genetic studies involving the respective chromosomes. The clones are available upon request.
