The loci for E-cadherin (uvomorulin, UVO) and N-cadherin (NCAD) have previously been assigned to human chromosomes 16q22.1 and 18, respectively (1, 2). We used cDNA clones for murine M-cadherin (Mcad) (3) and human P-cadherin (PCAD) for our mapping studies. The PCAD clone, isolated in a search for new members of the cadherin family, is a 2.4 kb cDNA clone, starting with bp 759 (according Shimoyama et al. (4)) and containing a poly A stretch at the 3' end. By Southern blot analysis using DNA from seven mouse x human somatic cell hybrids, the locus of the human homologue, MCAD, of mouse M-cadherin could be assigned to 16q24-qter, an interval to which APRT maps. Hybridization with the PCAD probe identifies a PCAD-like sequence, PCADLI, at 16q22.1, close to UVO, and an additional locus, PCADL2, at 16q24-qter. Very likely, PCADLl is the PCAD locus proper because the Pcad locus is closely linked to E-cadherin (uvomorulin, Um) and haptoglobin (Hp) on mouse chromosome 8(5). Using an interspecific backcross (6), we have mapped the mouse Mcad locus close to Aprt (no recombinants in 80 meioses), 7.5+/-2.9 cM distal to the E-cadherin (Um) locus.
