abstractText  Full text of MNL contribution: A MAJOR LOCUS CONTROLLING Beta-MANNOSIDASE IS LOCATED ON CHROMOSOME 3. Lundin L.-G., Hamar L. Dept. of Medical and Physiological Chemistry, BMC, Box 575, S-751 23 Uppsala, Sweden. A variation in the activity of Beta-mannosidase was first detected in the BXD series of RI strains. We have been able to confirm the presence of a major variation by using crosses between C57BL/6 and an inbred strain called CSB. This last strain is 1ikely to have descended from 101 X C3H mice. The C57BL/6 strain has high activities and the CSB and DBA/2 strains have ].ow activities in liver and kidney. There are very smal1 differences in spleen activity between B6 and D2. The activity groups, based on assays of liver and kidney homogenates, show little over1ap. There are associations between the mannosidase activities and the cdm locus in the BXD series and between mannosidase and the Adh-3 locus in the CSB X B6 back-cross material. Thus, there is a major locus controlling Beta-mannosidase in the distal part of chromosome 3 for which we suggest the gene symbol Bmn. It is still not clear whether we are dealing with a structural or a regulatory locus. There are correlations between kidney and liver activities but we have not yet been able to pinpoint any structural differences between the enzymes from B6 and D2 mice that might explain the difference in activity. The correlations are much weaker between activities in spleen on one hand and kidney and liver on the other hand. This is likely to be due to the effect on spleen activity of a second locus which causes the spleen activities in B6 and D2 to be almost equal. Furthermore, there are indications that this locus is close to Lsh on chromosome 1.
