Corticotropin-releasing factor (CRF) is the principal neuroregulator of the secretion of ACTH and plays an important role in coordinating the endocrine, autonomic and behavioral responses to stress. CRF exerts its effects by initially binding to a membrane-bound receptor. We have recently cloned a cDNA coding for the CRF receptor from a human corticotropic tumor by expression cloning. The CRF receptor is a member of G protein-coupled calcitonin/VIP/GRF receptor subfamily. The receptor expressed in COSM6 cells binds rat/human CRF with high affinity and specificity and is functionally coupled to adenylate cyclase. By southern analysis of DNAs prepared from a mapping panel of human/hamster and human/mouse hybrid cell lines (Coriell Cell Repositories), we report here the CRF receptor gene to be on human chromosome 17. In order to study the physiological function of the CRF receptor and the regulation of its gene expression, we have characterized the mouse CRF receptor gene. Using a DNA fragment covering the coding region of the rat CRF receptor as a probe, we screened a mouse phage genomic library. Several positive clones were obtained and the inserts were cut out by Not1 digestion and subsequently cloned into pBluescript. The genomic structure of the mouse CRF receptor was characterized by restriction mapping an partial DNA sequencing of these positive clones. The mouse CRF receptor gene comprises at least 12 exons, spanning more than 14 kb. We are currently characterizing the promoter region of the mouse CRF receptor gene. |
