Study of the mechanism of T-cell-mediated cytotoxicity have now given way to a search for molecules involved in this function. These molecules may be identified by detecting the corresponding messenger RNA transcripts. Such an approach may include, as a first step, the search for transcripts as specific as possible to cytotoxic T-cells; only secondarily can their actual relevance to cytotoxicity be investigated. Four distinct messenger RNA transcripts (CTLA-1, CTLA-2, CTLA-3, CTLA-4) were detected by a systematic screening of a substracted complementary DNA bank. The corresponding genes were then mapped, bu in situ hybridization, on metaphase spreads from a WMP male mouse whose entire chromosome complement, other than the 19 and X chromosomes, is associated in the form of multiple robertsonian translocations (kindly give by JL Guenet, Pasteur Institute, Paris). The CTLA-1 gene was thus localized in the D band of mouse chromosome 14, in a region to which the T-cell receptor alpha-chain gene (TCRA) has been mapped. The CTLA-2 and CTLA-3 genes were assigned respectively to the B band and the D band of mouse chromosome 13. The CTLA-4 gene was assigned to the C band of mouse chromosome 1.
