The cathepsins comprise a group of intracellular proteases which have been found in vertebrate and invertebrate tissues. The major cathepsin activities include a group of cysteine-dependent proteases, cathepsins B, H and L, which are structurally related to papain. Subcellular fractionation studies have shown that the mature, active forms of cathepsins are localized predominantly in the lysosome where they play and important role in regulating intracellular protein degradation and turnover. Cathepsin B and H recently have been cloned (Chan et al. Proc Natl Acad Sci USA 83:774, 1986). Using these cDNA's as hybridization probes, we report the chromosomal localization of CTSB and CTSH. Employing human-mouse somatic cell hybrids, we mapped CTSB to chromosome 8, and CTSH to the q11-qter region on chromosome 15. In situ hybridization further localized CTSB to 8p22 and CTSH to 15q24-25. |
