The chromogenic substrate alpha N-0-met (N-acetyl-L-methionine alpha-naphthyl ester, Schaller and von Deimling (1979) Analyt. Biochem. 93: 251-256) was applied for the electrophoretic demonstration of submandibular gland ester-proteases. Using disc electrophoresis and isoelectrofocusing in polyacrylamide gels, we compared the esterprotease patterns of C57BL/l0Sn with those of four other inbred strains (A/J, DBA/2J, NMRI, Sk/Cam) and with the wild stocks kl/oci (a substrain of M.m.molossinus, obtained from V. Chapman) and M.m. castaneus (J. Womack). We found genetic polymorphisms for protease A (Schenkein), protease E (a possibly new esterprotease) and the epidermal growth factor binding protein (Cohen). The results are being published in Comp. Biochem. Physiol. (Otto and von Deimling). We have now analyzed a total of 314 backcross progeny. We found that the three polymorphic enzymes are coded for on chromosome 7. The locus coding for protease A showed 21.4% recombinations with c and 42.8% recombinations with Es-8. No recombination was observed between the three esterproteases. However, because of the biochemical differences between protease A and protease E and the different distribution of alleles on the strains surveyed, separate structural genes should be responsible at least for these two esterproteases. We have classified both enzymes as serine proteases (EC 3.4.21) and suggest the gene symbols Prt-4 for protease A and Prt-5 for protease E. The two allelic variants we found at the Prt-4 locus have been designated Prt-4a (higher anodal mobility, DBA/2J) and Prt-4b (lower anodal mobility, C57BL/10Sn). The four different forms, controlled by Prt-5, are Prt-5a (highest IEP, above 8.5, kl/oci) Prt-5b (medium IEP, 8.5, C57BL/10Sn), Prt-5c (lowest IEP, 7.5, DBA/2J) and PrtÐ5d (no expression, Sk/Cam). Based upon our recombination data the proposed gene order is Es-8 - Prt-4/Prt-5 - c. The location of the esterprotease loci seems to be very near to L.Skow's Tam-1. Therefore it now appears that loci encoding for submandibular gland esterolytic proteases are clustered on chromosome 7. (Otto).
