abstractText | Full text of MNL contribution: 3. Detailed mapping of serum amyloid A complex (Saa) on Chr 7. Our previous analysis of serum amyloid A (SAA) gene restriction fragment variants showed that SAA related sequences map in a tight cluster closely linked to the Tam-1 cluster between Gpi-1 and p on Chr 7 (Taylor and Rowe, Mol. Gen. Genet., in press). No recombinants were detected between Tam-1 and Saa among 17 RI strains. We have examined 113 (C3H/ HeJ-pJ x IS/CamEi)Fl female x C3H/He.J-pJ male and 57 C3H/HeJ-pJ female x (C3H/HeJ-pJ x IS/CanEi)F1 female, backcross progeny in order to more precisely localize Saa. These progeny were bled and typed for Gpi-1. Recombinants between Gpi-1 and p were further typed for Pep-4, Tam-1, Prt-5, and Saa. The gene order and map distances in cM obtained are shown below: F1 females (N=113): Gpi-1/Pep4: 3.5+/-1.7; Pep-4/Tam-1, Prt-5: 4.4+/-1.9; Tam-1, Prt-5/Saa: 3.2+/-1.7; Saa/p: 6.5+/-2.3. F1 males (N=57): Gpi-1/Pep-4: 4.4+/-2.8; Pep-4/Tam-1, Prt-5: 6.6+/-3.3; Tam-1, Prt-5/Saa: 4.4+/-2.8; Saa/p: 2.2+/-2.0. Combined (N=170): Gpi-1/Pep-4: 3.8+/-1.5; Pep-4/Tam,1, Prt-5: 5.0+/-1.7; Tam-1, Prt-5/Saa: 3.4+/-1.4; Saa/p: 4.8+/-1.7. These distances and standard errors are adjusted to account for loss of information about a few mice. Two mice from Fl females that were (Tam-1, Prt-5) Ð p recombinants could not be typed for Saa, and two mice from F1 males that were Gpi-1 - p recombinants died before they could be tested further. Any double crossovers in the Gpi-1 p region would have been included in the parental classes. We have shown by restriction analysis of mice trisomic for the segment of Chr 7 inserted into the X in T(X;7)1Ct that the Saa complex is not within the insertion. Since ru-2, which is 3 cM proximal to p, is known to be included in the inserted segment, we conclude that the proximal Chr 7 breakpoint of T(X;7)1Ct is in the short interval between Saa and ru-2.
