abstractText  Full text of Abstract: cDNA Cloning of a Mouse IL-3 Receptor alpha Subunit. Takahiko Hara and Atsushi Miyajima. DNAX Research Institute, Palo Alto, CA 94304. We have recently deomstrated that the high affinity human IL-3 receptor is composed of an IL-3 specific alpha subunit which is shared among IL-3, GM-CSF and IL-5 receptors. In contrast to man, mouse has two genes, AIC2A and AIC2B, which are homologous to the human common Beta subunit. AIC2A binds mouse IL-3 (mlL-3) with low affinity (Kd-5-20 nM) and evidence indicates that it is a component of a high affinity IL-3R (Kd=100-500 pM). As we were not able to find a mlL-3R alpha gene by using the human alpha cDNA as a probe, we tried expression cloning. COS cells were transfected with small pools of clones from a cDNA library together with the AIC2A cDNA and subjected to high affinity mlL-3 binding assays. We obtained a cDNA (pSUT-1) encoding a protein that by itself binds mlL-3 with very low affinity (Kd=45 nM). Coexpression of the SUT-1 and the AIC2A cDNAs in COS cells conferred high affinity mlL-3 binding (Kd=270 pM). Further characterization of the SUT-1 protein and the reconstituted high affinity receptor is in progress.
