abstractText  Full text of MNL contribution: Research News: A cDNA from proximal human chromosome 15q maps near Mtv-1 on mouse chromosome 7. Several cDNA probes have been isolated from a human fetal brain library (Neve et al, Mol Brain Res 1:271-280, 1986; Tantravahi et al, Am J Med Genet 32: in press, 1989) via hybridization to the human chromosome 15 genomic probes (Donlon et al, PNAS 83:4408-4412,1986; Nicholls et al, Am J Med Genet 33: in press, 1989). The cDNA probe DN10 is highly conserved in at least man, monkey, mouse and chicken. *DN10 detects 12 fragments totalling 110 kb in HindIII digested human DNA. Data from somatic cell hybrids regionally map all fragments to human chromosome 15pter-15ql4. Deletion analysis utilizing DNAs from Prader-Willi syndrome patients refines the localization to proximal 15q. DN10 also detects multiple fragments in restriction enzyme digests of mouse DNA. Six Recombinant Inbred (RI) progenitor strains were tested and revealed restriction fragment length polymorphisms (RFLPs) with 10/12 enzymes. Four alleles were distinguishable with MspI : DN10a- AKR/J, C3H/HeJ ; DN10b - C57BL/6J, C57L/J ; DN10c- SJL/J; DN10d - DBA/2J. Strain distribution patterns (SDPs) were determined in 4 sets of RI strains using MspI and the data are summarized in the accompanying table. DN10 segregates with Mtv-1, Tam-1 and Xmv-33 on mouse chromosome 7. DN10 is approximately 1 cM from Mtv-1, with gene order Tam-1----Xmmv-1----DN10, Xmv3 3----Mtv-1----Mod-2.
