We have used DNA probes for Smst, the gene coding for somatostatin, and Ets-2, a protooncogene, in conjunction with Chinese hamster-mouse somatic cell hybrids segregating mouse chromosomes, to regionally map Smst and Ets-2 on mouse chromosome 16. Somatic cell hybrids containing either the proximal 4/5 or the distal 1/5 of mouse chromosome 16 as defined by the T(2;16)2811 reciprocal translocation were formed by fusing mouse peritoneal macrophages homozygous for the T28H translocation with Chinese hamster recipient cells. Southern blot analysis of DNA from these somatic cell hybrids with a 340 base pair BglII-SmaI probe isolated from a rat somatostatin cDNA revealcd a 25kb mouse Smst-specific EcoRI fragment only in hybrids containing the segment of chromosome 16 proximal to the T28H breakpoint. Conversely, Southern blot analysis with a 166 base pair HindIII-PstI probe derived from the avian erythroblastosis virus E26 revealed a 2.9kb mouse Ets-2-spccific PstI fragment only in those hybrids containing the segment of chromosome 16 distal to the T28H breakpoint. These findings place Smst proximal and Ets-2 distal to the T28H breakpoint on mouse chromosome 16. We have previously reported that mouse Sod-1 and Prgs, homologous to genes assigned to human chromosome 21, map distal to the T28H breakpoint on mouse chromosome 16. Since the human genes homologous to Smst and Ets-2 map to the long arms of human chromosome 3 and human chromosome 21 respectivelty, the results reported here provide further evidence for homology between human chromosome 21 and the distal tip of mouse chromosome 16.
