The mammalian ras c-onc family is composed of the Harvey, Kirsten, and N-ras genes. They encode 21kD GTP-binding proteins, associated with the plasma membrane, and may function in signal transduction in a manner akin to G proteins of the adenylate cyclase and transducin systems. A new transcribed ras gene, R-ras, was isolated from a human genomic library by low stringency screening with viral Ha-ras. The predicted 218 amino acid sequence shares 47% homology with human c-Ha-ras1, including several highly conserved domains. However, R-ras encodes an additional 26 NH2 terminal amino acids, and location of introns do not match Ha-, Ki-, or N-ras. A major 4.4 kb EcoRI R-ras fragment segregated with human chromosome 19 in human-mouse somatic cell hybrids. R-ras is thus unlinked to other known human ras genes. Mouse R-ras has 88% end 94% nucleotide and amino acid homologies respectively with human R-ras. A 6.7 kb HindIII mouse R-ras fragment was localized in mouse-Chinese hamster hybrids to chromosome 7; also the site of cHa-rasl. R-ras appears to be part of a conserved linkage group on human chromosome 19 and mouse chromosome 7 that includes GPI, PEPD, and LHB. The conserved linkage group including a-Ha-rasl on mouse chromosome 7 (and on human chromosome lip) appears to be more distal to the mouse chromosome 7 centromere than the R-ras linkage group. |
