abstractText | Full text of Abstract: A comparative clone map of the region containing the PKD1 gene in mouse. P.G. Olsson, C. Lohning, A.-M. Frischauf. Imperial Cancer Research Fund, London (UK) There is a conserved synteny group between human chromosomal region 16pl3.3 and mouse chromosome 17 (Himmelbauer et al., Genomics 13:35 (1992)) including the human polycystic kidney disease (PKD1), tuberous sclerosis 2 (TSC2) and cyclin F genes. The synteny group extends to the alpha-globin locus 1.7 Mb distal to the human PKDl gene, while the current most proximal markers are the mouse D17Lon2 and its human counterpart NK92. We have cloned the genomic mouse region containing the TSC2 and PKD1 genes in P1 and cosmid clones. The TSC2 and PKD1 genes are in close proximity and organized in tail to tail (3' to 3') fashion. The distance in between the poly-(A) signals of the two genes is 52 bp in man and 63 bp in mouse, including a 21-bp 90% conserved sequence close to the mouse poly-(A) signal. We have also isolated mouse cDNA clones containing homologues to several other genes in the region and mapped them to mouse chromosome 17. |
