abstractText | Full text of Mouse Genome contribution: Research News: 1. A New Imprinting Region in Band H1 of Mouse Chromosome 2. We have recently shown that neuronatin, Nnat, is maternally imprinted and lies between the breakpoints of T26H and T2Wa on mouse Chromosome (Chr) 2 (Kikyo et al., 1997, Developmental Biology, in press) and thus proximal to the imprinting region that is known to affect morphology, behaviour and viability of neonates (Cattanach and Kirk, 1985, Nature 315: 496-498). In this study, we have investigated how close Nnat lies to the distal Chr 2 imprinting region. Nnat was positioned in relation to other translocation breakpoints shown in the figure, by looking for its expression/repression in mice by RT-PCR. Nnat was expressed in the brain of newborn MatDp.dist2. T1Go mice that carry maternal duplication of the region distal to the breakpoint of T1Go in band H3. Nnat was also expressed in the brain of newborn MatDp.dist2. T1Sn mice that carry maternal duplication of the region distal to the TlSn breakpoint in and H1. These results place Nnat proximal to the breakpoint of TlSn in band H1 and this was verified by FISH analysis. We have also shown by linkage analysis that Nnat lies close to agouti, some alleles of which are influenced by parental inheritance (Michaud et al., 1994, Genes and Development 8: 1463-1472). In summary Nnat lies between the breakpoints of T26H and TlSn in band HI and defines a new imprinting region on Chr 2. This is approximately 18 Mb proximal to the previously identified region on distal Chr 2. (Williamson, Beechey, Ball, Dutton, Tease, Peters) There is a Figure on page 688. |
