description  Single-cell RNA sequencing was performed on injured and uninjured hindlimb tissue of Ddr2 slie/slie mice and littermate controls following 30% total body surface area burn and Achilles's tenotomy injury to determine the role of DDR2 in heterotopic ossification and aberrant differentiation during wound healing. Ddr2 slie/slie mice (n=2) and littermate controls (n=2) were placed under isoflurane anesthesia and provided subcutaneous buprenorphine analgesic prior to 30% total body surface area burn and concurrent left Achilles' tenotomy. Tissue from the left injury site, which includes the Achilles' tendon, from the tendon-muscular junction at the gastrocnemius muscle and calcaneal enthesis, and surrounding tissue posterior to the tibia, was dissected one week following injury. Right limb tissue was also harvested to serve as the contralateral limb control. Injured left limb tissue within each group was combined for processing, as were right limbs for each group. Tissue was digested and processed into a single-cell suspension for sequencing. Four samples were submitted for single-cell RNA sequencing including Ddr2 slie/slie injured tissue, Ddr2 slie/slie uninjured tissue, littermate control injured tissue, and littermate control uninjured tissue.
