description  Single-cell sequencing of diaphragms from control, SRSF1 knock-out, and SRSF2 knock-out mice revealed distinct cell types and highlighted the different roles of SRSF1 and SRSF2 in the development and function of skeletal muscle satellite cells. The diaphragms were isolated from one-day-old control, SRSF1 KO, and SRSF2 KO mice. The knockout of SRSF1 or SRSF2 was driven by Cre recombinase under the control of the MyoD1 promoter. To obtain a single-cell suspension, the diaphragms were fully digested with Dispase I and Collagenase type XI. The resulting single cells were then used for single-cell sequencing.
