description | The evolutionarily conserved serine 182 residue on RORgammat is phosphorylated. Here, we report that this residue is dispensable for thymic T cell development, but essential for maintaining proper balance of Th17-Treg populations in the colon. Single-cell RNA-seq (scRNA-seq) revealed that serine 182 residue on RORgammat is critical for restricting IL-1b-mediated Th17 activities and promoting anti-inflammation cytokine IL-10 production in Lymphoid tissue (LT)-like RORgammat+Foxp3+ regulatory T cells in inflamed tissues. Two independent biological replicates in each condition were performed. In wildtype mice, results from DSS treated colon were compared to no treatment conditions. Results from RORgtS182A mutant mice were compared to those from wildtype mice in no treatment and DSS treatment conditions separately. |
