| description | Protein kinase N2 (Pkn2 ) plays a critical role in embryonic heart development, but the earliest molecular defects resulting from its loss in cardiomyocytes are not well defined. To capture primary transcriptional changes preceding overt morphological abnormalities, we performed bulk RNA-seq on embryonic day 10.5 (E10.5) hearts from cardiomyocyte-specific Pkn2 knockout (cKO) and littermate control embryos. At this stage, mutant and control hearts are grossly indistinguishable, allowing unbiased identification of transcriptional alterations directly caused by Pkn2 deficiency. This dataset provides genome-wide gene expression profiles from four cKO and four control embryonic hearts and serves as a resource for understanding the early transcriptional consequences of Pkn2 loss during cardiac morphogenesis. Embryonic hearts were dissected at E10.5 from Pkn2 fl/fl ; XMLC2-Cre (cKO) and Pkn2 fl/fl ; Cre (control) littermates. Ventricular tissue from four biological replicates per group was used to generate bulk RNA-seq libraries. Total RNA was extracted using TRIzol, poly(A)-selected RNA libraries were constructed using the Illumina TruSeq RNA Library Prep Kit v2, and libraries were sequenced as 100 bp paired-end reads on an Illumina NovaSeq X Plus platform. |
