abstractText | Full text of Abstract: Abstracts of workshop presentations. Two RFLPs at the locus of alpha2-HS glycoprotein (AHSG) on chromosome 3 are detected with a 1.2 kb probe. Alpha2-HS glycoprotein, a negative acute phase plasma protein, presents a genetic polymorphism when studied by thin-layer isoelectric focusing (Boutin et al. Am J Hum Genet 37:1098-1105,1985). Linkage and somatic cell hybrid studies have localized the AIISG gene to chromosome 3q (Eiberg et al. Cytogenet Cell Genet (HGM7)37:461,1984; Cox and Francke, Hum Genet 70:109-115, 1985). We have isolated from a human cDNA library established in lambda gt11, using immunochemical techniques with specific antibodies to the protein, a 1.2kb fragment. Partial nucleotide sequence indicates that it corresponds to the coding sequence of the protein, beginning at a.a. No. 99 of the A chain (probe A.99.AHSG 3'). Hybridization of this probe with DNA fragments from several human-Chinese hamster somatic cell hybrids confirmed the localization of the gene to chromosome 3q. No additional hybridizing sequences were detected. The A.99.AHSG 3' probe was used to screen DNA digests from 10 normal blood donors and an additional 29 individuals were screencd to determine allele frequencies. Digestion with Sac I allowed the identification of a common two-allele polymorphism due to the absence (.42) or the presence (.58) of a restriction site, resulting in one band (15 kb) or two bands (11 kb and 3.8kb). Digestion with Pvu II detected 1.7 kb and 2.5 kb alleles (.95 and .05, respectively). No definite polymorphism could be detected with 10 additional restriction enzymes. Invariant 0.9 kb, 1.2 kb, 1.5 kb and 5.6 kb bands were detected in Taq I digests, but a ladder pattern (500 bp spacing) extending at least 4 kb from the 5.6 kb band was observed in partial Taq I digests. This suggests that the AHSG gene is located in close proximity to satellite DNA. HGM symbol: AHSG |
