Hybridization of DNA from panels of cow-rodent hybrid somatic cells with molecular probes has resulted in the syntenic assignment of thirty seven additional loci to the bovine gene map and the assignment of two additional syntenic groups to specific chromosomes. AT3 maps to syntenic group U1 with PGD and ENO1; KRTB to U3 (chr 5) with GAPD and LDHB; HEXA and FOS to U5 with PKM2 and NP; TYR to U7 with LDHA; ASL and GUSB to U8 with MDH2; DIA4 to U9 with GPI; SST to U10 with SOD1; RHO to U12 with ACY1; HOX1, MET, and COLlA2 to U13; LGB to U16 with ABL; NEFL to U18 with IFNA; CAT, CRYA2, and PTH to U19 (chr 15) with HBB; HOX2 and KRTA to U21 (chr 19) with GH; AMH, LCB and SPARC to U22; IL2 and IGL to U23 with ALDH2; PLAT to U14 with GSR; and MOS, MYC, and CA2 to U24 (chr 14) with TG. PEPA was asyntenic with all other groups. Gene symbols correspond to current nomenclature of homologous human genes. In addition, the bovine caseins, CASAS1, CASAS2, CASB, and CASK were found to be syntenic with PGM2 and PEPS on U15 and subsequently localized to bovine chr 6 by in situ hybridization. A combination of in situ and somatic cell analysis has localized CRYG of syntenic group U17 to chr 8. The syntenic map of the cow now consists of 118 loci on 26 autosomal syntenic groups and the X and Y chromosomes. Seven of the autosomal groups are assigned to specific chromosomes. Comparative maps of cattle, man, and mouse demonstrate extensive chromosomal conservation between the three mammalian groups, particularly between the chromosomes of cattle and man. |
