JMJ Derry, NC Lan, JC Shih, EA Barnard, PJ Barnard. MRC Molecular Neurobiology Unit, MRC Centre, Cambridge, CB2 2QH, England. Monoamine Oxidases A and B are important enzymes in the central nervous system and peripheral tissues in the degradation of neuroactive and vasoactive amines and in the oxidation of xenobiotics. We have mapped both loci to the mouse X chromosome by pedigree breakpoint analysis in a mouse interspecies backcross. The two genes have been mapped in over 60 backcross progeny relative to seven known loci. We have found that both genes map to the Cybb - Timp interval on the mouse X chromosome, giving the gene order: cen - Cybb - Maoa/Maob - Timp - Hprt - Rsvp - Cf-8 - mdx - Pgk-1 - tel In 5 out of 6 animals recombining in this interval Maoa and Maob show co-segregation with Timp, suggesting they lie close together and centromeric to Timp. Our data provide further evidence for the syntenic relationship of loci in the Cybb à Timp interval in mouse and man, and predict a human localization of Xp21.1-p11.21 for both genes. This is in agreement with human mapping data for MAOA. |
