| abstractText | Full text of Mouse Genome contribution: Research News: En-1 is not allelic with Dh. The homeobox-containing gene engrailed-1 (En-1) maps to the same region of chromosome 1 as the developmentally significant gene dominant hemimelia (Dh). The following mapping experiment shows that Dh is not a mutant allele of En-1. Mice carrying the chromosome 1 marker genes ln, Dh, Pep-3b and the DNA markers Emv-17b and En-ld were mated to inbred C57BL/Ws animals (+, +, Pep-3a, Emv-l7d and En-lb). F1 animals were mated to mice from the Harwell LIII stock that were homozygous for ln, +, Pep-3b, Emv-17b and En-1d. The linkage study was carried out in two stages. The first stage involved typing 563 offspring for ln, Dh and Pep-3. This produced 56 recombinants. Stage two involved analysis of 43 of these recombinants by Southern hybridisation with the DNA markers Emv-17 and En-1. This experimental design avoided the necessity for molecular analysis of a large number of mice by focusing on those that were known to be recombinant in the ln-Dh-Pep-3 region of chromosome 1. However, since only 43 of the 56 recombinant mice were included in stage two of the study, the effective total number of mice is correspondingly reduced to 432.
