abstractText | Full text of Abstract: Genetic Mapping of the Mouse X Chromosome in the Region Homologous to Human Xq26-Xq28. G.E. Herman, M.G. Darlison, and E.A. Barnard, Baylor College of Medicine, Houston, Texas and MRC Molecular Neurobiology Unit, Cambridge, England. While conservation of X-linked loci between man and the mouse is absolute, at least 5 major evolutional rearrangements between the X chromosomes of the two species have occurred. Data from other laboratories has suggested that loci mapping to human Xq26-Xq28 may represent a conserved linkage group on the mouse X. However, discrepancies with the human gene order were detected for the anonymous human probes C11(DXS144) and Stl4(DXS52) which detect murine loci designated DXPas6 and DXPas8, respectively. We have extended the analysis of DNA markers from this region of the mouse X chromosome using an interspecific cross involving C57BL/6JAw-J and Mus spretus. One hundred interspecific backcross progeny have been analyzed at 7 anchor loci spanning the mouse X chtomosome (Otc, Hprt, Cf-9, svp, Dmd, Pgk-1, and Ags). An additional 100 backcross progeny have been screened for recombination events between Cf-9 and Dmd; all recombinants were screened at the other anchor loci to identify any double crossovers. The cross was also designed to segregate the X-linkcd dominant mutation bare patches (bpa) which is probably homologous to human X-linked dominant chondrodysplasia punctata (CDPX2). Four single recombinants between Hprt and Cf-9 and 12 between Cf-9 and Dmd have been identified. These animals have been used to map other DNA probes from this region including anonymous human probes C11 and St14; human cDNA probes for Factor 8 and the red visual pigment gene (RCP); a bovine cDNA probe for the GABAA receptor alpha3 subunit; and a mouse genomic probe for the P3 locus which is approximately 70 kb distal to G6PD. Pedigree analysis of recombinant backcross progeny suggest a gene order of Otc-(Hprt, DXPas6)-(Cf-9)- Gabr-alpha3-(DXPas8, Bpa)-(Rsvp, Cf-8, P3)-Dmd. This order agrees with the order for the human loci mapping to Xq26-Xq28 and extends the conserved linkage in this region. It also suggests that the human CDPX2 locus maps in distal Xq27-Xq28 possibly between GABRalpha3 and RCP. |
