abstractText | Full text of Abstract: 27040. Assignment of 47 additional comparative anchor loci to the bovine synteny map. Womack JE, Dietz AB, Gallagher DS, Li L, Zhang N, Neibergs HL, Moll YD, Ryan AM. Department of Veterinary Pathobiology/Texas A&M University, College Station, TX 77843, U.S.A. The bovine synteny map consists of over 200 gene loci on all 29 autosomal syntenic groups plus the X and Y chromosomes. Most of the mapped loci are comparative anchor loci in that their homologs have been previously placed on the human and mouse maps. Utilizing our panels of hybrid cells, in combination with both Southern blotting and PCR amplification techniques, we have assigned an additional 47 anchor loci to the bovine map. These genes are CGA and ESR (U2); AC02 (U3); CHGA and IFG1R (U4); TCRA and HGMCR (U5); IGF2 (U7); PCKB (U8); HP and LHB (U9); C9 (U10); OT, ARVP, HCK, and PRIP (U11); INHBA, PGY3, TCRB, and TCRG (U13); D4S43, D4S10, QDPR, KIT, IGJ, ADH2 and IF (U15); POMC and CD8A (U16); FUCA1P (U17); IFNO and TP (U18); CD3E and CD3D (U19); LDLR, INSR, ADRB2R, CSF2, RPS14, PDGFRB, FGFA, and CSF1R (U22); FGB, FGG, and F11 (U23); F10 (U27); and YES1 (U28). Gene symbols are from latest HGM Workshop list of gene loci and homology of mapped bovine genes was critically tested. A bovine-mouse-human comparative map is presented which defines 35 evolutionary disruptions in the bovine-human map. The same anchor loci define 49 disruptions in the mouse-human map. These data fill previous gaps in the three-way comparative map and support the hypothesis that bovine-human chromosomal conservation exceeds that of human and mouse. |
