abstractText | Full text of Mouse Genome contribution: RESEARCH NEWS. INTERSTRAIN DIFFERENCE STUDY OF SUSCEPTIBILITY TO THIO TEPA, EXPERIMENT WITH RECOMBINANT STRAINS OF MICE. Clastogenicity of thioTEPA in mice of raising recombinant strains, from 101/H x C3H/Sn cross was studied. In summer 1993 15 strains have reached inbreeding F10. 5-6 sibs of each strain (females and males) aged 6-8 weeks were treated with mutagen (3 mg/kg, i/p). Chromosome preparations of bone marrow cells were made 24 hours after the treatment. The results are obtained on the base of 100 metaphases analized per animal. The strain susceptibility was evaluated as frequency of the cells with chromosome damages. The strains were divided into two unequal groups by this characteristic. The first group included 12 less susceptible strains, the average frequency of cells with chromosome aberrations being 33.5% (min = 27, 0 ± 4, 9; max = 39, 8 ± 4, 9) The second group included 3 highly susceptible strains - 54, 1% (1XC3-9 -51, l +/- 5, 0; 1XC3-17 - 51, 7 ± 5, 0; 1XC3-6 - 59, 5 ± 4, 9%). Similar division was found by the number of chromosome breakage per cell: 1, 15 in resistant and 2, 88 in susceptible strains. The difference between the groups was significant and similar to that found earlier for strains C3H and 101. Thus the present data confirm the hypothesis that mice of strain 101/H posess the gene increasing the susceptibility to chemical mutagenes (mut-1). Part of the group 1 strains is eliminated and the rest will be bred till F20 and then they will be studied in more detail once more. (A. Malashenko, T. Beskova) |
